LOABeads

LOABeads magnetic beads combine the high binding capacity of agarose with fast separations due to excellent super-paramagnetic behavior.

LOABeads are a flexible alternative for any bioseparation application, from microliter scale to liter scale. The magnetic agarose beads have the same properties that are valued in normal agarose, such as high binding capacity and minimal nonspecific interactions. At the same time the beads provide the simplicity and fast separations of magnetic beads, though at a larger scale than what has previously been available.

LOABeads magnetic beads are agarose beads imbued with magnetic material. With a diameter of approx. 100 µm and a dark color, the individual beads are clearly visible by eye, making collection simple. They do not form aggregates and are easily dispersed without use of detergents.

Employing the LOABeads beads together with Lab on a Bead’s coupling chemistry allows the introduction of affinity, IEX and hydrophobic ligands. This makes LOABeads suitable as a high-capacity and specificity media for purification of biomolecules using fast and easy magnetic separation techniques.

Coupling chemistry

Fast and easy coupling of proteins to surfaces at physiological conditions.

Lab on a Bead’s unique patented coupling chemistry enables fast and efficient covalent conjugation of amino (-NH2) containing ligands, such as proteins, and thiol (-SH) containing ligands to surfaces. All steps in the coupling are performed at physiological conditions and at room temperature with high yields and short reaction times. The use of water-based media eliminates the use of organic solvent and no hazardous reagents are needed. This makes Lab on a Bead’s coupling chemistry a more ecofriendly alternative compared to most of the traditionally used coupling techniques, such as the CNBr and NHS methods.

The technology is used in the production of LOABeads Protein A magnetic beads and is available for sale with the LOABeads AffiAmino beads. Affinity, IEX and hydrophobic ligands can be introduced, making a wide variety of bioseparations possible using magnetic beads.

Bench scale magnetic bioseparation

Lab on a Bead brings the simplicity and flexibility of magnetic beads to bioseparation on a liter scale.

By combining the LOABeads magnetic beads with the LOABeads MagSep magnetic separators, a new platform for laboratory scale bioseparation is formed. The beads will capture milligrams to grams of the target molecule from production volumes of up to several liters. Using the magnetic separators, washing and elution of the target protein can then be performed in standard flasks. By combining the LOABeads MagSep units, target proteins can be captured from a larger volume and then washed and eluted in a smaller sized vessel

LOABeads Protein A

High capacity protein A magnetic agarose beads provide rapid and simple purification of antibodies using magnetic separation techniques.

LOABeads Protein A have a binding capacity 60 mg human or rabbit IgG per ml settled magnetic beads. The black beads are clearly visible and easily attracted to external magnets, enabling distinct separation within seconds, which leads to minimal sample loss. The 4% agarose matrix offers minimal unspecific binding and makes the beads non-adherent, letting the beads be easily dispersed and resuspended without the use of detergents.

The purification setup can easily be scaled up or down to match antibody concentration and sample volumes. LOABeads Protein A work excellently for separations from microliter to liter scales using suitable magnetic separators, such as the LOABeads MagSep series.

LOABeads Protein A are intended for purification of antibodies from serum and ascites fluid samples, IgG serum depletion, and immunoprecipitation. The product is for research use only and not intended for human use or for use in diagnostic applications.

LOABeads AffiAmino

Activated magnetic beads for coupling of amino containing ligands.

LOABeads AffiAmino are surface activated magnetic agarose beads employing Lab on a Bead’s patented coupling chemistry. Any amino containing ligand, e.g. proteins and antibodies can be covalently bound to the beads using a simple process. All steps are performed at physiological conditions and room temperature, without using any hazardous reagents or organic solvents. After conjugation of the ligand, the magnetic beads can be used for affinity purification and immunoprecipitation.

The binding capacity of LOABeads AffiAmino is 0.5–1 mg of human IgG per ml bead suspension (5–10 mg per ml settled beads), 0.5–0.75 mg Protein A per ml bead suspension (5–7.5 mg per ml settled beads) and similar amounts of other proteins. The bond is very stable (6 ppm leakage of Protein A) and the beads can be stored with bound ligand.

The black beads are clearly visible and easily attracted to external magnets, enabling distinct separation within seconds, which leads to minimal sample loss. The 4% agarose matrix offers minimal unspecific binding and makes the beads non-adherent, letting the beads be easily dispersed and resuspended without the use of detergents.

LOABeads MabBind A

Magnetic agarose beads optimized for efficient purification of IgG from cell cultures.

LOABeads MabBind A are protein A coupled magnetic agarose beads specifically developed for the purification of monoclonal IgG antibodies from cell culture harvests, such as from hybridoma and CHO cell cultures. The high specificity and high capacity of LOABeads MabBind A ensures that a very pure antibody is obtained with a high yield, even when antibody capture is performed directly in the cell culture medium.

The black beads are clearly visible and easily attracted to external magnets, enabling distinct separation within seconds, which leads to minimal sample loss. The 4% agarose matrix offers negligible unspecific binding and makes the beads non-adherent. This lets them be easily dispersed and resuspended without the use of detergents.

LOABeads MabBind A are ideal to use in many applications of magnetic bead affinity chromatography (MBAC). For example in lab-scale purification of antibodies to be used in R&D and diagnostics, and for characterizations of candidate biopharmaceutical IgG antibodies. The quantity of beads can readily be scaled up or down and regardless of scale, the same conditions will provide equal yield and purity.

For analytical and small-scale preparative work, as well as method scouting, samples can be handled in standard 15 and 50 ml centrifuge tubes using the LOABeads Magsep 15/50 separator. In the LOABeads MagSep 500 unit, 30 ml of settled beads in a 500 ml sample can be handled, giving a maximum adsorbent capacity of 900 mg IgG.

LOABeads MagSep

Durable magnetic separators for volumes between milliliter and liter.

The LOABeads MagSep series of laboratory magnetic separators have been optimized for use with LOABeads magnetic beads and are ideal for most bioseparation applications. They are designed for durability with a lacquered plastic shell and nickel plated neodymium magnets placed safely inside. The separators fit standard test tubes and are available in sizes from 15 ml to 500 ml.

The combination of LOABeads magnetic particles and LOABeads MagSep is a simple biomagnetic separation platform that requires no special training to use. The separators require very little maintenance and several batches can be processed in parallel using the same unit, making the running costs very low.

Handling

The handling of LOABeads in the LOABeads MagSep magnetic separators follow the same simple procedure regardless of the size.

  • The tube or flask containing well-suspended LOABeads is placed in the separator.
  • The magnetic beads are immediately attracted by the surrounding magnets and within seconds or minutes, depending on the scale, the separation is complete.
  • The liquid is removed using a pipet while the magnetic beads are held by the surrounding magnets.
  • The tube or flask is removed from the separator and the magnetic beads are resuspended in the appropriate buffer.
